Carnosine protects against Abeta42-induced neurotoxicity in differentiated rat PC12 cells
Laboratory studyOther animals
- Design
- Laboratory study
- Dose used in the study
- 5 mM carnosine for 18 hours before 5 microM Abeta42 for 24 hours
- What was measured
- Cell viability, glutamate release and surface NMDA receptor expression in differentiated PC12 cells exposed to Abeta42, with histamine receptor antagonists or a histidine decarboxylase inhibitor added before carnosine.
What the authors reported
Carnosine pretreatment ameliorated the Abeta42-induced loss of viability. The protection was reversed by the H3 antagonists thioperamide and clobenpropit but not by H1 or H2 antagonists or by alpha-fluoromethylhistidine. Carnosine reduced glutamate release and increased surface NMDA receptor expression in the presence of Abeta42. The authors conclude the mechanism is independent of the carnosine-histidine-histamine pathway and may involve glutamate release and NMDA receptor trafficking.
