Effect of polaprezinc on impaired healing of chronic gastric ulcers in adjuvant-induced arthritic rats: role of insulin-like growth factors (IGF)-1

Kato S, Tanaka A, Ogawa Y, et al.. Medical Science Monitor, 2001; 7(1):20-25.

Controlled trialOther animalsWith zinc (as polaprezinc)

This study tested carnosine together with zinc (as polaprezinc). Its results cannot be assigned to carnosine alone. More combination studies.

Design
Controlled trial
Dose used in the study
polaprezinc 3 to 10 mg/kg by mouth twice daily; omeprazole 30 mg/kg by mouth once daily; recombinant human IGF-1 30 micrograms/kg subcutaneously twice daily
Duration
14 days of treatment, starting 3 days after ulceration
What was measured
Healing of thermally cauterised gastric ulcers and mucosal IGF-1 mRNA in arthritic versus normal Dark Agouti rats, with polaprezinc, omeprazole or recombinant IGF-1.

What the authors reported

Ulcer healing was delayed in arthritic rats on days 10 and 17, and the rise in IGF-1 mRNA in ulcerated mucosa was attenuated. Polaprezinc accelerated healing in normal and arthritic rats in a dose-dependent manner, more so in arthritic rats. Omeprazole promoted healing in both groups. Recombinant IGF-1 promoted healing in arthritic rats only. The authors suggest polaprezinc acts probably through an increase in IGF-1 production.

Limits of this study

Animal numbers not stated in abstract. Rodents lack serum carnosinase, so oral results do not carry over to people. Full text not read.
