Analysis of an H1 receptor-mediated, zinc-potentiated vasoconstrictor action of the histidyl dipeptide carnosine in rabbit saphenous vein

O'Dowd A, Miller DJ.. British Journal of Pharmacology, 1998; 125(6):1272-1280.

Laboratory studyOther animals

Design
Laboratory study
Dose used in the study
carnosine in rings up to 10 mM; binding assay with zinc at 10, 30 and 80 microM
What was measured
Contraction of isolated rabbit saphenous vein rings and [3H]-mepyramine binding to H1 sites in guinea-pig cerebellar membranes.

What the authors reported

Mepyramine antagonised carnosine-induced contractures (Schild pA2 7.97) and histamine-induced contractures (pA2 8.48). Histamine contractures were smaller (73%) and less well sustained than carnosine contractures. Serotonergic antagonists methiothepin and ketanserin also antagonised both, which the authors read as coincidental H1 inhibition. With zinc present, carnosine inhibited [3H]-mepyramine binding to guinea-pig cerebellar membranes (log IC50 -2.78, -3.93 and -4.64 at 10, 30 and 80 microM zinc, corrected for zinc's own inhibition). Carnosine also evoked an H2-mediated relaxation in the presence of mepyramine. The authors conclude carnosine can act at the smooth muscle H1 receptor.
