- What was measured
- Fluorescence imaging of intracellular calcium and pH in cardiac myocytes during cytoplasmic acidification by acetate or photolytic proton release, plus proton uncaging into buffer mixtures in vitro.
What the authors reported
Raising intracellular H+ evoked a rise in intracellular Ca2+ independent of sarcolemmal Ca2+ influx or release from mitochondria, sarcoplasmic reticulum or acidic stores. In vitro, proton uncaging showed Ca2+ unloading from proteins, histidyl dipeptides (for example carnosine) and ATP can underlie the rise. A local acid microdomain recruited Ca2+, reduced by magnesium overload or ATP depletion. The authors conclude histidyl dipeptides and ATP underlie spatial Ca2+/H+ coupling by exchange on common buffer sites.
Limits of this study
